    一用途

    葉綠素含量測(cè)量?jī)x廣泛應(yīng)用于植物生理生態(tài)研究,特別是對(duì)傳統(tǒng)吸收法測(cè)量困難的小葉片或樣品的葉綠素含量進(jìn)行測(cè)量,例如針葉、發(fā)育未*的水稻、生于巖石上的絲狀藻、地衣、草坪草、仙人掌、龍舌蘭屬植物、菠蘿、擬南芥、果實(shí)、苔蘚、葉莖、葉柄。

    二原理

    盡管實(shí)踐已經(jīng)證明了吸收技術(shù)測(cè)量葉綠素含量的有效性,但該方法對(duì)樣品的要求較高,使其應(yīng)用范圍受到限制。而樣品本身的葉綠素?zé)晒鈩t不受樣品厚度、均一性等的影響,可以指示葉綠素的含量。根據(jù)這個(gè)原理,Gitelson等比較了葉片的吸收和反射對(duì)葉綠素?zé)晒獾挠绊?,發(fā)現(xiàn)在735nm和700nm處的比值與葉片的反射相關(guān)性最高,并且當(dāng)樣品葉綠素含量在41mg·m-2到675mg·m-2時(shí),F(xiàn)735/F700的比值與其葉綠素含量間存在著顯著的線性關(guān)系(R2≥0.95),與傳統(tǒng)的吸收性測(cè)量方法相比(測(cè)量范圍≤300mg·m-2),提高了葉綠素含量的線性關(guān)系范圍。

    根據(jù)Gitelson的研究結(jié)果,CCM-300使用具有15nm半峰寬的460nm二極管用于熒光激發(fā),提高了紅色熒光波長(zhǎng),避開了大約685nm的發(fā)射熒光峰(葉綠素可重新吸收,降低測(cè)量精度)。儀器使用在735nm和700nm的葉綠素發(fā)射熒光的比率(F735/F700)進(jìn)行葉綠素含量的測(cè)量,讀數(shù)為F735/F700的比值以及根據(jù)Gitelson方程計(jì)算的葉綠素含量。

技術(shù)參數(shù)

Ø 重復(fù)性:依賴于信號(hào)強(qiáng)度,對(duì)信號(hào)強(qiáng)度低的樣品,建議使用多點(diǎn)測(cè)量的平均值,對(duì)信號(hào)強(qiáng)度高的樣品,差異通常為±0.03或更低;。

Ø 光源:LED 460 nm藍(lán)光二極管,半峰寬15 nm

 

Ø 探頭:兩個(gè)固態(tài)高感傳感器,提供濾光片。同時(shí)提供兩個(gè)波長(zhǎng)探頭(700nm~710nm,730nm~740nm);
